摘要
目的:探讨C反应蛋白(CRP)对内皮细胞株ECV304细胞释放前列环素(PG I2)、纤溶酶原激活物抑制剂-1(PAI-1)的影响。方法:用不同浓度的CRP与ECV304细胞培养不同时间后,用ELISA法检测培养液中PG I2的稳定代谢产物6-keto-PGF1α、PAI-1的含量。结果:用CRP作用ECV304细胞后,PAI-1的浓度随CRP浓度增加和CRP作用时间延长而呈递增趋势,而6-keto-PGF1α水平在培养12小时内上升,12小时后其水平下降,并随CRP浓度增加而减少。结论:CRP可促进内皮细胞释放PAI-1,抑制PG I2的分泌,从而促使血管痉挛、血栓形成和急性冠脉综合征的发生发展。
Objectlve:To observe the effect of CRP on se - cretions of PGI2 ,PAI - 1 in ECV304 cells. Methods:Incubation of ECV304 ceils with different concentration of CRP for different time, and then measure the concentration of 6 -keto -PGF1α ,PAI - 1 in cultural fluid by ELISA. Results. Incubation of ECV304 cells with CRP results in a time - and dose - dependent increase in secreted PAI - 1 and a significant inhibition of PGI2 release. Conclusions: CRP induces the production of PAI- 1 and quenches the production of PGI2 in ECV304 cells and thus has implications for angiospasm ,thrombosis and ACS.
