高效液相色谱法测定动物组织中抗生素的残留量

Determination of residual antibiotics in animal tissues by high-performance liquid chromatography

提出了猪肌肉组织中四环素、土霉素和氯霉素的高效液相色谱测定方法。肉样中的四环素和土霉素用 EDTA-磷酸混合液提取、经三氯乙酸沉淀蛋白质后,上清液用 Sep-Pak C_(18)柱富集、净化后得测定液。肉样中的氯霉素用磷酸盐缓冲液提取,三氯乙酸沉淀蛋白质后,上清液直接作为测定液。本法采用 NucIeosil C_(18)色谱柱,磷酸氢二钠缓冲液-EDTA-乙腈和磷酸二氢钠缓冲液-乙腈为流动相系统。四环素、土霉素、氯霉素的平均回收率分别为74.5%～79.2%、71.1%～79.5%、71.0%～72.2%;相对标准偏差(RSD)分别为3.0%～6.8%、4.2%～7.9%、3.5%～3.7%。

The determination of tetracycline,oxytetracycline and chloramphenicol in porcine muscle by high per- formance liquid chromatography (HPLC) is presented.Tetracycline and oxytetracycline were extracted from homogenized meat with a mixture of EDTA-phosphoric acid and deproteinized with trichloroacetic acid.The supernatant was concentrated and extracted through a sep-pak C_(18) cartridge and the elute was analyzed by HPLC.Chloramphenieol was extracted from homogenized meat with a phosphate buffer and deproteinized with trichloroacetic acid.After filtration,the supernatant was analyzed by HPLC.Nucleosil C_(18)(7μm) 4× 250 mm column was used,and the mobile phase were disodium monohydrogenorthophosphate buffer-ED- TA-acetonitrile for tetracycline and oxytetracycline,and sodium dihydrogenorthophosphate buffer-acetoni- trile.for chloramphenicol.The average recoveries were 74.5%～79.2%,71.1%～79.5% and 71.0%～ 72.2%,and the relative standard deviation (RSD) were 3.0%～6.8%,4.2%～7.9% and 3.5%～3.7%, respectively for tetracycline,oxytetracycline and chloramphenicol.
