摘要
目的对葎草多糖的提取、分离纯化和单糖的组成进行了分析。方法葎草经热水提取乙醇沉淀得多糖组品(HSW),经脱蛋白、透析、DEAE-纤维素柱和SephadexG-100层析进行纯化,纯化多糖通过色谱等方法进行分析。结果粗多糖主要含HSW1、HSW2、HSW3、HSW4四个组分。结论各组分经高效凝胶色谱、薄层层析、气相色谱等分析得知:HSW1由葡萄糖(Glu)、半乳糖(GaD组成,其摩尔比为1.00;0.21,HSW2~3均由鼠李糖(Rha)、阿拉伯糖(Ara)、甘露糖(Man)、葡萄糖(Glu)、半乳糖(Gal),其摩尔比分别是0.73:0.99:0.69:0.80:1.00及0.51:0,31:0.48:1.00:0.45,HSW4主要由甘露糖(Man)、葡萄糖(Glu)、半乳糖(GaD,其摩尔比1:0.42,为葎草多糖更深的研究打下了基础。
OBJECTIVE To purify and analysis the polysaccharides from Hurnulus scandens (Lour.)Merr. METHODS The crude polysaccharides(HSW) were obtained by extracting hot water and precipitating with alcohol. The cleared HSW were purified by DEAE-cellulose,Sephadex G-100 column,and it were analyzed by chromatography. RESULTS The purified four proteins (HSW1-4) were removed from crude polysaccharides. CONCLUSION Analysis by. HPGC, TLC and GC,The molecular weights of HSW1 - 4 were 2.0 × 10^4, 5. 6 × 10^4, 6. 8 X× 10^5,and 1. 4 × 10^6, respectively. The chemical components and the molar ratio showed follows :HSW1 : Glu : Gal=1.00 : 0. 21;HSW2'S and HSW3'S component were. Rha,Ara,Man,Glu,Gal,the molar ratios showed 0. 73 : 0.99 : 0. 69 : 0. 80 : 1.00 and 0. 51 : 0. 31 : 0.48 : 1.00 : 0.45;All these work could establish foundation for the further study of the Hurnulus Polysaccharides.
