摘要
DNA修复相关蛋白Rad51是真核细胞进行同源重组修复的关键酶。在肿瘤细胞中Rad51基因可出现异常表达,引起重组修复失控,基因组不稳定,因此与肿瘤的发生、发展密切相关。同时Rad51蛋白水平在肿瘤细胞中的升高,可导致肿瘤对放化疗产生抵抗作用。以Rad51为肿瘤治疗靶点,降低其在肿瘤细胞中的表达水平,可提高细胞对放化疗的敏感性,这一思路有可能发展为新的肿瘤治疗手段。
In eukaryotic cells, Rad51 protein plays a key role in the pathway of homologous recombination. Abnormal expression of RadS1 gene in tumor cells can lead to the disturbance of homologous recombination process and genetic instability. It is clear therefore that the abnormal expression associated with Rad51 is closely correlative to the development and growth of tumors. The high level of Rad51 protein in tumor cells may result in the resistance of chemotherapy and radiotherapy. Therapy targetting Rad51 gene can reduce the expression of Rad51 gene expression and increase effects of radiotherapy and chemotherapy. Based on this theory, new methods may be developed on tumor therapy.
