摘要
目的:建立高效液相色谱法测定甲磺酸酚妥拉明的含量。方法:采用MACHEREY—NAGEL氰基色谱柱,以0.05mol/L磷酸二氢钾溶液(取磷酸二氢钾6.8g,加水1000ml溶解后,用10%磷酸溶液调节pH值至3.0)-乙腈(4:1)为流动相,流速为1ml/min,检测波长为278nm,进样量20μl。结果:甲磺酸酚妥拉明的线性范围为20~80μg/ml,r=0.9999(n=7),平均回收率为99.49%,RSD=0.38%。结论:本方法快速、简便、重现性好、专属性强,可用于控制甲磺酸酚妥拉明的内在质量。
Objective:To establish an HPLC method for determination of Phentolamine Mesylate. Method:The chromatography was carried out with MACHEREY-NAGEL CN column and mobile phase of 0. 05mol/L potassium phosphate monobasic (weighed 6.8g Potassium phosphate monobasic,added water to 1000ml,adjusted to pH 3.0 with 10% phosphoric acid)-acetonitrile (4 : 1) with detective wavelength at 278nm. The flow rate of the mobile phase was 1ml/min. Results:The calibration curve was linear in the range of 20-80μg/ml, r = 0. 9999 (n= 7). The average recovery was 99.49% ,RSD= 0. 38%. Conclusion :This method is rapid,simple,well repeatable and specific. It can be used as a method of quality control.
