摘要 目的:总结并分析单纯性肥胖分子生物学机制,并初步探讨了运动减肥的分子生物学机制。资料来源:应用计算机检索Medline1995-01/2004-12关于肥胖及运动减肥的文章。检索词“exercise,obesity,leptin,uncouplingprotein”,并限定文章的语种类为English。同时利用计算机检索中国期刊全文数据库1994-01/2004-12的相关文章,限定文章语言种类为中文,检索词“运动,肥胖,瘦素,解耦联蛋白”。资料选择:对资料进行初审,纳入标准:①关于单纯性肥胖中重要的中枢调控因子。②运动对肥胖的影响。排除标准:重复性研究。资料提炼:共收集到符合上述要求的文献113篇,对内容重复及与主题关系较远的文章进行筛除,24篇符合纳入标准。资料综合:肥胖是食欲和能量调节紊乱引起的疾病,与遗传、膳食结构、环境等多种因素有关,其中基因是主要决定因素。leptin和Orexin目前被确认是和肥胖进程密切相关的调控因子。解耦联蛋白基因作为肥胖的候选基因,它的表达与肥胖的进程密切相关。运动减肥是通过对肥胖基因的影响来实现的。结论:leptin抵抗对肥胖影响深远。运动可以提高leptin的效应,改善leptin抵抗,进而减去体脂。运动对leptin通路具体环节的影响以及解耦联蛋白等基因的影响还有待于进一步研究。 OBJECTIVE: To summarize and analyze molecular biology mechanism of simple obesity, and probe into primarily molecular biological mechanism of exercise dieting. DATA SOURCES: A computer-based online search of Medline database was undertaken to identify articles about the molecular biological mechanism of obesity and exercise dieting published in English from January 1995 to December 2004 by using the key words of "exercise, obesity,leptin,uncoupling protein". Meanwhile, related articles about the above topic published between January 1994 and December 2004 were searched for in China Journal Full-text Database (CJFD) with the key words of "exercise, obesity, leptin, uncoupling protein" in Chinese. STUDY SELECTION: The literatures were selected primarily. Inclusive criteria were (1)researches of the main alerting genes of nerve center in simple obesity, (2)researches of the effect of exercise on obesity. Exclusive criteria: repetitive researches were excluded. DATA EXTRACTION: There were 113 literatures in accordance with the above-mentioned criteria, and 24 were associated with inclusive criteria. Articles of repetitive content and far away from the topic were excluded. DATA SYNTHESIS: Obesity was caused by disorderly regulating of appetite and energy and related to such factors as gene, food structure and environment and so on. Gene was the main crucial factor. Both leptin and orexin were taken as main obese gene to regulate and control the process of obesity. Genes of uncoupling protein were correlation to the obesity process as candidate gene. And it suggested that obese gene were affected in training. CONCLUSION: Leptin resistance plays a critical role in obesity. The effect of leptin is elevated and leptin resistance is improved in training in order to subduce body lipid. It needs further studies that how exercise affects the signal conducting of leptin and the gene of uncoupling protein.
