黄花倒水莲皂苷C抑制ox-LDL诱导的LOX-1的表达 被引量:10

摘要 目的:观察黄花倒水莲皂苷C抑制氧化型低密度脂蛋白(oxidizedlowdensitylipopprotein,ox-LDL)诱导的人脐静脉内皮细胞血凝素样氧化型低密度脂蛋白受体(oxidizedlowdesitylipoproteinrecep-tor-1,LOX-1)表达的作用。方法:培养人脐静脉内皮细胞株(HUVEC-12),随机分为:对照组、ox-LDL(50mg/L)组和氧化型低密度脂蛋白(50mg/L)+黄花倒水莲皂苷C(1,3,10μmol/L)组,通过RT-PCR和Westernblot蛋白印迹分析检测LOX-1mRNA和蛋白的表达。结果:ox-LDL上调LOX-1mRNA和蛋白的表达,黄花倒水莲皂苷C明显抑制ox-LDL诱导的LOX-1mRNA和蛋白的表达,且呈浓度依赖性。结论:黄花倒水莲皂苷C明显抑制ox-LDL诱导的内皮细胞LOX-1mRNA和蛋白的表达。 Objective To investigate the effect of reinioside C ( RC ) on the expression of lectin-like oxidized low density lipoprotein receptor (LOX)-1 mRNA and LOX-1 protein induced by ox- idized low density lipoprotein (ox-LDL) in cultured human umbilical vein endothelial cells (HUVEC ). Methods presence of RC ( were examined by icantly raised the ent. Conclusion duced by ox-LDL HUVECs were cultured with ox-LDL (50 mg/L) for 24 h in the absence or 1, 3, and 10 μmol/L). The expressions of LOX-1 mRNA and LOX-1 protein RT-PCR and Western-blot. Results Incubation with ox-LDL (50 mg/L) signifexpression of LOX-1 mRNA and LOX-1 protein, which was concentration-dependent.Conclusion RC can inhibit the increased expression of LOX-1 mRNA and LOX-1 protein induced by ox-LDL in HUVECs.
