葛根芩连汤配伍葛根素在兔体内药动学的研究

目的:测定葛根芩连汤煎剂和葛根单煎剂中葛根素于兔血中的含量,分析配伍对其体内过程的影响。方法:采用高效液相色谱法,测定不同时间点兔血浆葛根素的含量,W inNon lin软件计算药动学参数。结果:葛根芩连汤全方及葛根单煎剂中葛根素在兔体内过程均符合二室开放模型,主要药动学参数分别为,AUC(单)=70.66±3.54 m in.μg/m l,AUC(全)=231.87±17.06m in.μg/m l,Tm ax(单)=23.93±0.32m in,Tm ax(全)=27.789±0.078m in,Cm ax(单)=0.29±0.01μg/m l,Cm ax(全)=0.36±0.08μg/m。l结论:葛根芩连汤配伍能提高葛根素的达峰浓度及生物利用度,具有促进葛根素吸收的作用。

To investigate the pharmacokinetics of various combinations of pueraria in Gegenqinlian decoction. Methods： The concentrations of puerarin in Gegenqinlian decoction in plasma were determined by HPLC. The WinNonlin program was used to calculate the pharmacikinetic parameters. Result： The puerarin in gegen and Gegenqinlian decoction could be described as an open two-compartment model. Its main pharmacokinetic parameters were as follows ： AUC （only） = 70.66 ± 3.54min·μg/ml, AUC （all） = 231.87 ±17.06min·μg/ml, Tmax （ only ） = 23.93±0.32 min, Tmax （all） = 27. 789 ±0.078min, Cmax （only） = 0.29± 0.01μg/ml, Cmax （ all ） = 0.36± 0.08μg/ml. Conclusion： Gegenqinlian decoction can improve the Cmax and bioavailability of puerain, the decoction can promote the absorb of puerain.
