摘要
目的:建立人血清中左卡尼汀药物浓度测定的反相高效液相色谱法。方法:采用反向色谱柱Hypersil ODS C18,流动相为磷酸盐缓冲液(用磷酸调pH值为3.0)-甲醇(3:1);流速1.0 mL·min-1;检测波长225 nm。结果:左卡尼汀在1~100μg·mL-1范围内线性关系良好,相关系数r=0.999 9。结论:该方法灵敏、准确、简单、快速,可用于临床左卡尼汀血药浓度监测和药动学研究。
Objective:To develop a RP-HPLC method for the determination of L-carnitine in hu- man serum. Methods:The RP-HPLC condition was consisted of a Hypersil ODS C18 column, a mobile phase of phosphate buffer( pH = 3.0)-methanol(3:1 )with a flow rate at 1.0 mL-min-2, and the detection wavelength at 225 nm. Results: The calibrated linear curve of L-carnitine concentration was in the range of 1 - 100μg·mL^-1(r = 0. 999 9). Conclusion:This sensitive, reliable and rapid RP-HPLC method could be used in the pharmacokinetic study of L-carnitine.
