缺血预处理与心肌缺血/再灌注细胞凋亡及NF-κB表达的关系 被引量:2

摘要 目的:研究缺血预处理对心肌缺血/再灌注细胞凋亡及NF-κB表达的影响。方法:健康SD大鼠30只随机分为3组:对照组,缺血/再灌注组,缺血预处理组,每组10只。利用心肌缺血/再灌注模型,采用原位末端标记(TUNEL)法测定凋亡细胞,采用W estern-b lot法测定NF-κB的表达。结果:缺血再灌注组心肌细胞的凋亡百分数和NF-κB的表达均显著增加,与对照组相比有显著差异;而缺血预处理组与缺血/再灌注组相比,其凋亡心肌细胞百分数和NF-κB的表达均显著减少。结论:缺血预处理抑制心肌细胞的凋亡的作用机制与其抑制NF-κB的表达有关。 Objective: To investigate the effect of ischemia pretreatment on myocardium ischemia /reperfusion apoptosis and the expression of NF-κB. Methods: 30 healthy SD rats were divided into 3 groups randomly: control group, ischemia/reperfusion group and ischemia pretreatment group. Apoptosis cell was estimated by TUNEL staining and the expression of NF-κB was assayed by Westernblot method, Results: Myocardium cell apoptosis percentage and the expression of NF-κB both increased obviously in ischemia/reperfusion group and the difference was significant compared with control group, In ischemia pretreatment group, apoptosis percentage and the expression of NF-κB decreased significantty compared with ischemia/reperfusion group. Conclusion: The mechanism of ischemia pretreatment inhibiting myocardium apoptosis is associated with its inhibition the expression of NF-κB.
