摘要
目的:为建立全面评价罗汉果的质量的方法。方法:采用超声强化水提取法进行样品前处理,用罗汉果苷Ⅴ作为参照物进行指标成分峰的定位,用HPLC对罗汉果水提取物进行分离,比较了水—甲醇、冰醋酸水溶液—甲醇、磷酸水溶液—甲酸、水-乙睛、磷酸水溶液—乙腈、三氟乙酸水溶液—乙腈6种体系的等度、梯度洗脱效果。结果:实验表明在上述6种流动相体系中,以三氟乙酸水溶液—乙腈的洗脱效果最好,色谱条件:梯度洗脱、SinochromODS-BP柱、检测波长为205nm,流速0.8ml/min,柱温25℃。结论:采用HPLC技术,可以得到分离度和重现性均较好的罗汉果水提取物的HPLC色谱图。
Objective:To establish a method for quality evaluation of mogroside. Method: The strengthened ultrasonie extraetion technology was used for the sample pretreatment. Active component peak was located by the reference compound mogrosideV. Mogroside aqueous extracts were separated by HPLC. The effects of isoeratie elution and gradient elution for six moving phase systems, i.e. water - methanol, aeetie aeid - methanol, phosphorie aeid - methanoie aeid, water - methyl eyanide as well as phosphorie aeid - methyl eyanide and trifluoroaeetie aeid - methyl eyanide were eompared. Result: The better separated conditions are using trifluoroaeetie acid - methyl cyanide as moving phases of the gradient elution under the deteetod wavelength of 205 nm, flow rate of 0.8mL/min and eolumn temperature of 25℃ using a eolumn of Sinoehrom ODS - BP. Conclusion: The ehromatogram of mogroside aqueous extracts with better resolution and repeatability can be obtained by HPLC,
