抑癌基因PTEN及环氧化酶-2在大肠癌中的表达及其意义

摘要 目的:通过检测抑癌基因PTEN和环氧化酶-2(COX-2)在大肠癌中的表达,探讨其在大肠癌中的意义。方法:应用SP法免疫组化方法检测10例正常大肠黏膜、80例大肠癌组织中PTEN和COX-2的表达。结果:正常大肠黏膜中PTEN为强阳性表达,在大肠癌中有41.2%为阴性或弱阳性表达,明显低于正常组(P<0.01);COX-2在正常大肠黏膜中80%为阴性表达,而在大肠癌中阳性率为72.5%,显著高于正常组的表达(P<0.01);在大肠癌中PTEN和COX-2存在负相关(P<0.05)。结论:抑癌基因PTEN和COX-2在大肠癌的发生发展过程中起着重要的作用。 Objective To detect the expression of tumor suppressor gene PTEN and cyclooxygenase 2 (COX2) in colorectal cancer, and analyze their significance. Methods SP immunohistochemical method was used to detect the expression of PTEN and COX-2 in 10 cases of normal colorectal membrane and 80 cases of colorectal cancer. Results The expression of PTEN in all of the normal colorectal membrane were positive, and expression of PTEN in 41.2% cases of colorectal cancer were low or negative, which was much lower than the normal group(P〈0.01). The negative rate of COX-2 in normal group was 80%, while in colorectal cancer, the positive rate was 72.5%, which was significantly higher than normal group (P〈0.01). A negative correlation was observed between the expression of PTEN and COX- 2. Conclusion Our data suggest that PTEN and COX-2 play an important role in the development of colorectal cancer.
