摘要
从土壤样品分离、实验室保藏菌种及引进菌种共200多个菌株中,经纸层析定向筛选,获得20株具有显著产α-半乳糖苷酶活性特性的菌株。进一步以R15#菌株为出发菌株,经60Co诱变处理,及紫外—亚硝基胍复合诱变,得到产α-半乳糖苷酶性能稳定、活力较高的变异菌株3个,其产酶活力达231 U/g以上,平均比出发菌株提高190.9%。其中RM48#菌株经连续传代7代,产酶性能未出现较大的变异,具有较好的遗传稳定性。该菌株经鉴定为黑曲霉变种(Aspergillus nigerv.Tiegh)。
The activities of the α-galactosidase of about 200 strains were examined by paper chromatography. Twenty strains which obviously indicated α-galactosidase activities were selected. The R15 # strain was done original strain and mutagenized by ^60Co and UV-NTG complex mutagenests. Three mutant strains of high α-galactosidase production and production stability were selected. The enzymatic activities were improved about 190.9% and reached 231 U/g. The RM48# strain has stable enzyme production after subculture 7 times and was identified as Aspergillus niger v. Tiegh.
