摘要
目的通过测定急性肺损伤(acute lung injury,ALI)血浆及肺泡灌洗液中尾加压素Ⅱ(urotensinⅡ,UⅡ)的含量,来探讨尾加压素受体拮抗剂(UⅡreceptor antagonist,URA)对ALI中UⅡ的影响。方法Sprague Dawley(SD)大鼠28只,随机分为4组,每组7只:生理盐水对照组(A组);另三组以URA干预油酸型ALI动物模型,分别编为B、C、D组。3h后4个组均抽动脉血作血气分析,A组、B组抽动脉血后随即经心脏取血各2ml待测,C组、D组分别于12h、24h各取同量心脏血待测。各组均于心脏取血后活杀并取肺,以右肺作支气管肺泡灌洗(bronchoalveolar lavage,BAL),灌洗液(bronchoalveolar lavagefluid,BALF)离心后留取上清液,与心脏血分离的血浆一起在-80℃冻存待测。结果血气分析显示,B、C、D组血氧分压明显低于A组。A、B、C、D组血浆UⅡ质量浓度分别为(118·1±9·8)pg/ml、(133·6±8·9)pg/ml、(132·0±9·8)pg/ml、(114·0±9·1)pg/ml;相应各组BALF中UⅡ质量浓度分别为(10·1±1·4)pg/ml、(21·5±3·5)pg/ml、(63·1±6·5)pg/ml、(79·0±8·8)pg/ml。ALI大鼠血浆中UⅡ浓度进行性降低,而BALF中UⅡ进行性升高,差异具有统计学意义。结论UⅡ可能通过活化URA参与ALI的发病过程,并可能对ALI具有某种保护作用。
Objective To investigate the effects of urotensin Ⅱ (UⅡ) receptor antagonist (URA) on UⅡ in plasma and bronchoalveolar lavage fluid (BALF) in rats with acute lung injury (ALl). Methods Twenty eight Sprague Dawley (SD) rats were randomly divided into four groups: saline control group (group A), the other three groups (group B, group C, group D) . Rats were injected with oleic acid and URA to produce ALl models. Three hours after injection, arterial blood was drawn for blood gas analysis in all rats. Immediately after this, blood of heart was collected in group A and group B. Then blood of heart were collected after 12 hours in group C and after 24 hours in group D. Rats were killed when blood of heart was drawn. Bronchoalveolar lavage (BAL) with saline was performed in the right lung. BALF was centrifugated and the upper fluid was put to the EP tube. Plasma was separated from the blood of heart. Both plasma and BALF were kept at - 80℃ to be determined. Results Compared with group A, arterial pressure of oxygen was significantly decreased in the group B, group C, group D ( P〈 0.01) . U Ⅱ in plasma of group A to group D were (118.1±9.8) pg/ml, (133.6±8.9) pg/ml, (132.03±9.8) pg/ml, (114.0±9.1) pg/ml, respectively. UⅡ of group B, group C, group D in plasma were significantly decreased ( P 〈 0.05). U Ⅱ in BALF was significantly increased (P〈0.O1) in group A to group D, with concentration (10.1± 1.4) pg/ml, (21.5±3.5) pg/ml, (63.1±6.5) pg/ ml, (79.0 ± 8.8) pg/ml, respectively. Conclusion U Ⅱ may participate in the development of ALl, and URA maybe have some protection effect on ALI.
