摘要
目的探讨松萝酸钠对成纤维细胞的作用及其促伤口愈合的作用机制。方法建立体外细胞划伤模型,加入不同浓度的松萝酸钠(0.34,0.68,1.37,2.74μmol.L-1),观察不同作用时间(4,8,24 h)后,药物对细胞迁移行为的影响;采用ELISA法测定其分泌转化生长因子-β1(TGF-β1)和表皮生长因子(EGF)的变化规律。结果松萝酸钠对成纤维细胞迁移有促进作用;可促进成纤维细胞分泌TGF-β1和EGF,且随着药物浓度的增大而增强。结论松萝酸钠可通过促进成纤维细胞分泌TGF-β1和EGF,加速细胞迁移。
OBJECTIVE To investigate the mechanism of enhancing wound healing by sodium usnic acid(SUA). METHODS The in vitro cell scratch wound model was set up to observe the effect of SUA with different concentrations(0.34, 0.68, 1.37 and 2,74μmol· L^-1) and time(4,8 and 24 h)on cell migration;The levels of transforming growth factor-β1 (TGF-β1)and epidermal growth factor( EGF) were determined by enzyme-linked immunoadsorbent assay(ELIAS), RESULTS SUA accelerated cell migration and increased the secretion of TGF-β1 and EGF by fibroblasts, the action was reinforced with the increasing concentration of SUA. CONCLUSION The results indicate that SUA increased the secretion of TGF-β1and EGF, and accelerated the cell migration.
