摘要 目的检测胎盘组织中TGF-β1(transform ing growth factor-beta1,TGF-β1)及其受体TGFβRⅡ(TGF-beta receptorⅡ)的表达,探求TGF-β1在子痫前期发病中的作用。方法用半定量逆转录聚合酶链反应(RT-PCR)技术对10例正常妊娠组和26例子痫前期组(包括12例轻度和14例重度)胎盘组织中TGF-β1、TGFβRⅡmRNA的表达水平进行检测,并通过紫外凝胶图像分析进行定量分析。结果重度子痫前期组TGF-β1和TGFβRⅡmRNA的表达量均升高,与轻度子痫前期组及正常妊娠组相比有明显意义(P<0.05)。结论TGF-β1、TGFβRⅡ在转录水平就已经上调,它们的异常表达它们可能与子痫前期的发病有着重大的关系。 Objective: To detect the mRNA expression of transforming growth factorbetα1 (TGF- β1 ) and its receptor (TGF β RⅡ) in placentas in order to evaluate the role of TGF - β1 in preeclampsia. Methods : The semi - quantitative method of reverse transcription polymerase chain reaction (RT- PCR) was employed to investigate the mRNA expression levels of TGF - β1 , TGF β RⅡ in placenta tissue of 10 cases of normal term pregnant women and 26 cases of women with preeclmpsia ( 12 cases of mild preeclampsia and 14 cases of severe preeclampsia), the mean density was determined by image analysis technique. Results: Compared with the mild preeclampsia group and the normal group, the mRNA expression levels of TGF -β and TGF β RⅡ of severe preeclampsia group was higher ( P 〈0. 05). Conclusion The expression of TGF β1 and TGF β RⅡ was already changed at the level of transcription, the abnormal expression of TGF - β1and TGF Ⅱ RⅡ maybe play an important role in the pathogenic causes of preeclampsia.
