苦参药材中4种生物碱的含量测定 被引量:10

摘要 目的建立反相液相色谱法测定苦参药材中4种生物碱的含量测定方法。方法使用hypersil BDS(150mm×4.6mm,5μm)色谱柱,流动相为甲醇-0.1%氨水梯度洗脱,检测波长210nm。结果苦参碱在0.25~2.00μg(r=0.9997),槐定碱在0.28~2.24μg(r=0.9995),氧化苦参碱在0.244-1.953μg(r=0.9991),氧化槐果碱在0.262-2.096μg(r=0.9992)范围呈良好的线性关系,回归方程分别为y=2602.6x-72.031,y=1558.8x+17.381,y=1755.8x+162.59,y=2219.55x-21.119,加样回收率分别为99.24%,98.31%,101.28%,100.59%,RSD分别为1.248%,1.517%,1.412%,1.638%。结论该法简便,快速,专属性好,可用于苦参药材中生物碱的含量测定。 Objective To develop an HPLC method to determine the content of 4 alkaloids in Sophora flavescens Ait. Methods Hypersil BDS( 150 mm × 4.6 mm,5 um)phase column was used with mobile phase methanol- 0.1% ammonia water gradient elution ,the wavelength of determination was 210 nm. Results The linear concentration range of matrine ,sophoridine,oxymatrine and oxysophocarpine were 0.25 - 2.00 μg( r = 0.999 7 ) ,0.28 - 2. 24μg ( r = 0. 999 5 ) ,0. 244 - 1. 953 μg ( r = 0. 999 1 ) ,0. 262 - 2. 096μg( r = 0. 999 2 ), regression equation . were :y = 2 602.6x - 72.031 ,y = 1 558.8x + 17.381 ,y = 1 755.8x + 162.59 ,y = 2219.55x - 21.119, average recovery were 99.24% ,98.31% , 101.28% , 100.59% ,RSD were 1. 248% , 1. 517%, 1. 412%, 1. 638%, respectively. Conclusion This method is simple,rapid, sensitive, specific, and can be used for determination of the content of alkaloids in Sophoraflavescens Ait .
