摘要
目的探讨药物诱导SD大鼠大肠癌模型血清PpIX含量动力学变化规律。方法45只成功诱导大肠癌的SD大鼠分3组:A组ALA尾静脉注射;B组DFO腹腔注射半小时后ALA尾静脉注射;C组等量生理盐水尾静脉注射。各组用药后分别在1、2、4、6和12h经股动脉抽血,取其血清用高效液像色谱-荧光检测法测定PpIX含量。结果A、B两组大鼠血清内PpIX含量明显高于对照(C)组,B组PpIX含量明显高于A组(P<0.01);在0 ̄2h内血清PpIX的积聚率B组明显高于A组(P<0.05),在4 ̄6h血清PpIX的清除率B组明显高于A组(P<0.05)。结论DFO与ALA联合诱导,可使大肠癌SD大鼠体内PpIX迅速大量积聚并快速清除。
[Objective] To explore the dynamic changing nile of the content of Protoporphyrin IX (PpIX) in serum in colorectal cancer model in SD rats induced by drugs. [Methods] 45 SD rats with colorectal cancer induced by drugs were divided into three groups. Group A were injected with 5-ALA through tail veins. Group B were injected with 5-ALA through tail veins half an hour after celiac injection of DFO. Group C were injected with equal physiological saline through tail veins. Blood collection through femoral artery was performed in each group 1 h, 2 h, 4 h, 6 h, 12 h after drug injection, respectively. The content of PpIX in serum was determined by High Performance Liquid Chromatography (HPLC)-fluorescence detection method. [Results] The content of PpIX in serum in group A and group B was significantly higher than that in group C (control group). The content of PplX in serum in group B was significantly higher than that in group A (P 〈0.01). The accumulation rate of serum PpIx in 0-2 h in group B was significantly higher than that in group A (P 〈0.05). The clear-up rate of serum PpIx in 4-6 h in group B was significantly higher than that in group A (P 〈0.05). [Conclusions] PpIx can be accumulated and eliminated quickly in SD rats with colorectal cancer through induction by combination of DFO and ALA.
