胸腰段脊髓火器贯通伤后Bcl-2基因的早期表达

摘要 目的 建立家猪胸腰段脊髓火器贯通伤模型和改良Allen打击伤后全瘫模型,观察伤后Bcl-2基因的早期表达。方法 实验动物随机分为3组,其中火器贯通伤组(G组)9只,在全麻状态下制作胸腰段(L1~L2)脊髓火器伤模型;打击伤组(A组)9只,L1节段脊髓行改良Allen打击;对照组(C组)2只,仅予麻醉不致伤。于伤后1、3、6小时和距伤点不同距离处取材,进行Bcl-2蛋白的免疫组化染色。结果 两组脊髓损伤后6小时出现Bcl-2蛋白的表达增加,A组仅在伤点和近伤段处变化,而G组波及中伤段及远段伤。结论 细胞凋亡基因Bcl-2在脊髓损伤中的表达有一定的时空性,在脊髓损伤后6小时,出现Bcl-2基因蛋白表达增加。G组波及范围较A组更为广泛。 Objective To establish the models of thoracolumbar spinal cord gunsshot injury,and to investigate the early expression of Bcl-2 gene. Methods Twenty healthy pigs were randomly divided into 3 groups: gunshot injury group ( C group,n =9) ,the Allen's method induced injury group( A group,n =9) ,and the control group (C group, n = 2 ). Specimens were obtained 1,3,6h after injury. Histological changes were observed along with immuno-histochemistry of Bcl-2. Results The level of expression of protein of Bcl-2 gene increased 6h after injury, which could be observed at injury site and the proximal injury site in the group A,and at middle and distal injury sites in the group C. Conclusion The changes of Bcl-2 protein has timely and spatial characteristics in relation to the severity of SCI. Levels of protein expression of Bcl-2 gene increased 6h after injury. However,the injury areas of C group was more extensive than that of the group A.
