致敏和哮喘小鼠腹腔和支气管肺泡灌洗液巨噬细胞数量和功能表达 被引量:2

Expressions and Functions of Macrophage of Abdominal Cavity and Bronchalveolar Lavage Fluid in Sensitized and Asthmatic Mice

摘要 目的探讨抗原对巨噬细胞(Mφ)表达和功能的影响。方法用卵蛋白(OVA)腹腔注射和雾化吸入复制致敏和哮喘小鼠模型。在不同时间段收集腹腔冲洗液、支气管肺泡灌洗液(BALF),进行Mφ计数;采用流式细胞术和酶联免疫吸附法(ELISA)测定腹腔Mφ主要组织相容性复合体(MHC)Ⅰa抗原(MHC-Ⅰa)和培养上清液肿瘤坏死因子-α(TNF-α)的表达。结果腹腔Mφ(×106/mL),BALF细胞总数、Mφ数(×105/mL),致敏组小鼠致敏后2、5、7 d分别为7.59±1.27、2.01±0.61、1.66±0.52;19.43±6.37、2.87±0.50、1.84±0.40;7.68±2.28、1.40±0.45、0.99±0.38,哮喘组小鼠激发后2、57、d,分别为16.62±5.56、3.63±0.79、2.55±0.72;15.34±4.22、5.03±0.56、2.99±0.42;17.10±2.04、5.05±0.75、3.15±0.37。致敏组、哮喘组明显高于对照组(P<0.05)。致敏组、哮喘组腹腔MφMHC-Ⅰa和培养上清液TNF-α(μg/L)表达分别为44.09%5、9.28%;55.42±0.62,71.93±5.28,明显高于对照组(P<0.01)。结论致敏和哮喘小鼠腹腔冲洗液、BALF Mφ表达明显增加。MφMHC-Ⅰa和培养上清液TNF-α的表达明显增加。 Objective To explore the effects of antigen on expressions and functions of macrophages. Methods Mice were ,sensitized and challenged with ovalgumin(OVA) by abdominal injection and inhalation to establish an allergic and asthmatic model, Macrophages of the broncholveolar lavage fluid(BALF) and abdominal cavity were collected and quantity changes in different times were observed, Major histoeompability complex(MHC) I a expression of abdominal cavity macrophages were determined by flow cytometry, The levels of tumor necrosis factor-α(TNF-α) of abdominal cavity macrophage culture supernate were detected with enzyme linked immunosorbent assay (ELISA), Results After challenging, the second, fifth, seventh day, abdominal cavity rnacrophages( × 10^6/mL), total BALF cells( × 10^5/ mL)and alveolar rnacrophages( × 10^5/mL)in allergic group were 7.59 ± 1.27,2.01 ± 0.61,1.66 ± 0.52; 19.43 ± 6.37,2.87 ± 0.50, 1.84±0.40;7.68 ± 2.28,1.40 ± 0.45,0.99 ± 0.38, respectively. In asthmatic group, they were 16.62 ± 5.56, 3.63 ± 0.79,2.55 ± 0.72; 15.34 ± 4.22,5.03 ± 0.56,2.99 ± 0.42 ; 17.10 ± 2.04, 5.05 ± 0.75,3.15 ± 0.37, respectively, They were more than those of control group ( P 〈 0.05 ), MHC - I a expressions and TNF - α ( μg/L ) levels of culture supernates of abdominal cavity macrophages in allergic and asthmatic groups were 44.09 %, 59.28 % ; 55.42 ± 0.62,71.93 ± 5.28, As compared with control group, they markedly increased ( P 〈 0.01 ), Conclusions The expressions of macrophages of abdominal cavity and BALF in allergic and asthmatic group markedly increase, The expressions of MHC - I a and TNF- α of macrophages markedly increase.
