摘要
用大田软海绵酸与人IgG的偶联物OA-IgG作为免疫原,免疫BALB/c小鼠,通过常规细胞融合技术将免疫脾细胞与小鼠骨髓瘤细胞系SP2/0融合,用间接竞争ELISA方法对融合细胞所分泌抗体的特异性进行筛选。经过3次克隆,获得1株可稳定分泌抗OA的杂交瘤细胞株3C5,其染色体数为50±2,所分泌抗体属IgG1亚类,相对分子质量是159390,抗体亲和常数为9.8×108L/mol,滴度达2.56×106。
Spleen cells from mice hyperimmunized with a human IgG-okadaic acid N,N-dicyclohexyl carbodiimide conjugate were fused with SP2/0 myeloma cells. One hybridoma cell line was identified that secretes IgG1 monoclonal antibody, designated 3C5, against okdacid acid, after being subcloned for 3 cycles by indirect ELISA. It's average number of the chromosome was 50± 2. The molecular weight, the affinity constant and the titer of the McAb was 159 390, 9.8 × 10^8 L/mol and 2.56 ×10^6 respectively.
