高效液相色谱法测定血浆中消旋卡多曲的浓度及其药动学

HPLC determination and pharmacokinetics study of racecadotril in human plasma

目的：建立高效液相色谱法测定血浆中消旋卡多曲活性代谢物thiorphan（TP）的浓度。方法：血浆样品通过固相萃取技术处理后，采用Waters sunfire C18色谱柱（150mm×4．6mm．5μm）．以0．02mol·L^-1。磷酸二氢钾溶液（用磷酸调至pH2．6）-乙腈（74：26，V／V）为流动相；检测波长为210．进行样品测定。结果：血药浓度线形范围为0．05～4mg·L^-1（r=0．9999）。最低检测浓度为0．05mg·L^-1。提取回收率为93．50％～98．20％。方法学回收率为92．80％～％．60％．日内及日间RSD〈8．365％。结论：该方法灵敏．快速，准确.

OBJECTIVE An HPLC method was developed for determination of active metabolite TP of racecadotril in the plasma of human. METHODS The raberazole was extracted by extracting technology firmly from plasma and the applied column was Waters sunfire C18 5 m（ 150 mm × 4. 6 mm）. The mobile phase consisted of 0. 02mol· L ^-1 dihydrogen phosphate buffer solution （pH 2.6 adjusted with phosphric acid）-acetonitrile（74： 26）. Raberazole was detected by UV adsorption at 210 nrn. RESULTS The linear range of blood concentration was 0.05-4 g mL ^-1 （r= 0. 999 9） ,the limit of quantization was 0. 05 g·mL extraction recovery of raberazole was 93. 50%-98. 20%, the recovery of the method was 92.80% -96. 60%, the RSD of intraday and inter-day were RSD〈8. 365%. CONCLUSION The method is sensitive,fast and accurate.
