摘要
目的:建立人血浆中罗红霉素浓度的HPLC测定方法。方法:采用反相高效液相色谱法检测人血浆中罗红霉素的浓度:Kromasil C18色谱柱(瑞典,4.6mm×150mm,5μm),流动相:30mmol·L^-1醋酸铵(含1%的三乙胺.磷酸调pH值至7.8):乙腈。49:51,流速为1.0mL·min^-1.进样量40μL.柱温40℃,检测波长220nm。结果:标准曲线线性范围为0.25~32.0mg·ml^-1(r=0.9990,n=8)。血浆中罗红霉素的定量下限为0.25mg·mL^-1.日内RSD为5.05~6.47%,日间RSD为4.53~7.82%,萃取回收率为64.58~83.30%。结论:该方法简单,快速,重现性好.适用于人血浆中罗红霉素浓度的检测。
OBJECTIVE To establish a method for determining roxithromycin in human plasma by high performance liquid chromatography. METHODS Roxithromycin was successfully determined by reversed-phase high-performance liquid chromatography. Kromasil C18 column(150 mm×4. 6 mm,5 μm) was used. The mobile phase consisted of 30 mmol·L^-1 ammonium acetate solution containing( 1 % triethylamine, phosphoric acid, pH = 7. 8)-acetonitrile (49:51). All separation were at 40℃. and the flow rate was 1. 0 mL·min^-1 , Injection volume was 40 ILl.. The peak of roxithromycin was monitored to set at UV 220nm. RESULTS The linear range of roxithromycin was 0. 25-32. 0(g · mL^-1 (r= 0. 9990,n = 8). The detection limit was 0. 25 mg· mL^-1. The RSD of intra-day tests were 5.05-6. 47% and that of inter-day tests were 4. 53-7. 82%. The recovery of roxithromycin was 64. 58-83. 3%. CONCLUSION The method is simple, rapid and producible. It is suitable for determination of roxithromycin in human plasma.
