摘要
用聚合酶链反应法检测宫颈癌中人乳头瘤病毒16型转化基因E_7,检出阳性率为10/22.其中4例进行PCR-SSCP分析发现该基因与标准对照株存在明显变异。核苷酸序列分析证实有两处发生碱基突变。研究提示,PCR方法能快速检测宫颈癌中人乳头瘤病毒的转化基因,可作为宫颈癌的辅助诊断方法。SSCP分析能发现病毒株间小变异的基因片段。
he E_7 gene of human paillomavirus type 16 in cervical cancer was amplified by polyrnerase chain reactionand the positive rate of Ergene amplified is 10/22.The PCR products was analysed by a single-strandconformation polymorphism (PCR-SSCP)and the altered mobilities of amplified DNA fragroLent were observed inall the four cases analysed when compared with the E_7 gene insert of HPV16 plasmid.Two base mutations areconfirmed by nucleotide sequencing.This result suggested that the PCR can delect the transformation gene E7 ofHPV16 in cervical cancer rapidly and the PCR-SSCP can find variant fragments of a gene between the differentstrains of HPV.
