摘要
为研制一种 EB 病毒(EBV)基因探针,从鼻咽癌患者的活检组织、鼻咽分泌物和唾液中分离出 EBV,并从病毒中提取 DNA,克隆后用[α-^(32)P]dCTP 标记,制成[α-^(32)P]EBV-DNA 探针。再以患者活检标本、白细胞标本进行斑点杂交,用放射自显影法显示结果。临床试用表明:该探针诊断鼻咽癌的特异性强,灵敏度高,阳性率为92%(80/87),特异性为94.1%以上。这为鼻咽癌及与 EBV 有关疾病的诊断提供了一项检测方法,对上述疾病的流行病学调查及肿瘤基因学研究也有一定意义。
PURPOSE To prepare a EBV gene probe used for detecting nasopharyngeal carcinoma (NPC).METHODS EB virus was isolated from saliva and nasopharygeal secretion of the patients with NPC.EBV-DNA probe was prepared using[α-^(32)P] dCTP.Dot hybridization was performed with biopsy tissue sample and white cells of the patients.The results were expressed by autoradiography.RESULTS Clinical application showed that specificity and sensitivity of this probe were high for the diagnosis of NPC,reached about 94.1% and 92%(80/87) respectively.CONCLUSIONS It provided an important diagnostic method for the NPC and EB virus related diseases and also for the epidemiological study and tumor gene investigation.
