摘要
本文研究了两种不同的连接方法在野油菜黄单胞菌(Xanthomonascampestris)xanA基因与pUCm-T载体连接中的连接效果,并进行了转化实验进行鉴定。实验结果表明,xanA基因与pUCm-T载体的适宜连接条件为16℃连接过夜。连接体系中,xanA基因与pUCm-T载体的体积比为4:1。
Two methods for gene xanA to ligate with pUCm-T vector were discussed in this thesis. And the transformations were also done for testing. It showed that the better ligation condition was ligated at 1CC for overnight. In the ligation system, the volume ratio of gene xanA to pUCm-T vector was 4:1.
