外周血单个核细胞在乙肝相关性肾炎发病机制中的作用研究 被引量:4

摘要 目的通过观察外周血单个核细胞(PBMC)对体外培养的肾小管上皮细胞凋亡的影响,探讨PBMC在乙肝相关性肾炎发病机制中的作用。方法用流式细胞仪检测HBVDNA阳性血清与人近端肾小管上皮细胞(HK-2)培养时HK-2凋亡率作为对照,分别观察健康人及HBVDNA阳性患者PBMC对HK-2凋亡的影响。结果健康人PBMC组HK-2凋亡率明显低于对照组和HBVDNA阳性患者PBMC组(P<0.01)。结论健康人PBMC能抑制HK-2凋亡,而HBVDNA阳性患者PBMC此抑制作用降低,提示HBVDNA阳性患者PBMCs功能缺陷,可能是乙型肝炎相关肾炎(HBV-GN)的发病机制之一。 Objective To investigate the effects of peripheral blood mononuclear cells (PBMC) on apoptosis of renal tubular epithelial cells in vitro and study the role of PBMC in the mechanism of hepatitis B virus associated glomerulonephritis (HBV-GN). Methods The normal human kidney proximal tubular cell line (HK-2) were cultured in the presence of the HBV DNA positive serum, and then influenced by the PBMCs from healthy individual and HBV DNA positive patients, respectively. After 72 hours incubation, the apoptosis of the HK-2 was assayed by flow cyto-metry. Results The PBMCs of healthy individual rapidly induced a significant decrease of the apoptosis rate of HK-2, compared with the control group and the PBMCs from HBV DNA positive patients group ( P 〈 0. 01 ). Conclusion HBV persistent infection may decrease the inhibitory effects of PBMCs on HK-2 apoptosis, which may be an important mechanism of HBV-GN.
