摘要
目的预测p55抗原表位及蛋白性质。方法运用生物信息学方法推测p55抗原表位及蛋白性质,分析预测结果。结果p55蛋白存在多个潜在的抗原表位,T细胞表位按预测积分高低排列位于141-158,54-68,123-137,349-364,397-414位氨基酸残基;B细胞表位在37-41,74-79,111-119,148-161,199-210,240-250,256-266,295-314,322-360位氨基酸残基等部位。P55重组融合蛋白为可溶性蛋白的概率是92%。结论p55抗原表位可能主要存在于148-161、295-320、337-356氨基酸序列区域内或其附近。该预测p55抗原表位可以有目的地克隆重组基因片段,为研究高效的表位疫苗奠定基础。
By means of bioinformatics analysis, the antigenic epitopes and properties of the p55 protein in Pneumocystis carinii were analyzed. It was shown that multiple potential antigenic epitopes were predicted to be present in p 55 protein of P. carinii. Based on the predicted scores, the T cell epitopes were located at amino acid 141-185, 54-68, 123-137, 349-364 and 397-414 residues respectively; while the B cell epitopes located at amino acid 37-41, 74-79, 111-119, 148-161, 199-210, 240- 250, 256-266, 295-314 and 322-360 residues respectively. The probability of solubility of the p55 recombinant fusion protein was 92%. FrOm these predictive values, it is clear that the main antigenic determinants of p55 protein may be adjacent to amino acid 148-161, 295-320 and 337-356 residues. These results would be helpful for the purposely cloning of the recombinant gene fragments and can provide a basis for the vaccine development for P. carinli.
