摘要
目的:观察顺铂、氧化苦参碱对人肝癌细胞株QGY端粒酶活性的影响,进一步探讨两种药物的可能作用机制。方法:采用人肝癌细胞株QGY作为研究对象,MTT法检测不同浓度和作用时间的药物对细胞增殖的抑制作用,以及对QGY细胞粘附能力的影响;流式细胞仪分析细胞周期的分布及凋亡;PCR-ELISA法检测端粒酶活性的变化。结果:两种药物均具有抗QGY细胞增殖、阻滞细胞周期、诱导凋亡及抑制端粒酶活性的作用。结论:对端粒酶活性的抑制可能是两种药物发挥抗肿瘤作用的机制之一。
Objective:To investigate the anti-cancer mechanism of cisplatin, oxymatrine by observing their effects on telomerase activity. Methods:The inhibition of proliferation in QGY cells were estimated by MTT-test. Distribution of cell cycle and apoptosis were analysed using flow cytometry, The changes of telomerase activity were detected by PCR-ELISA method, Results:These two drugs could inhibit the proliferation of QGY cells, block the cell cycle, induce its apoptosis and inhibit the activity of telomerase.Conclusion: The inhibition of telomerase activity may be one of the mechanisms for the anti-cancer effect of the two drugs.
