摘要
目的观察串珠素对bFGF诱导的心肌微血管内皮细胞增殖的影响。方法分离并培养SD大鼠心肌微血管内皮细胞,经VIII因子相关抗原抗体鉴定和常规处理后,随机分为对照组、bFGF组、缺氧组、缺氧+bFGF组、PD98059组和串珠素阻断组,以[3H]-TdR掺入法测定内皮细胞增殖反应。结果缺氧+bFGF组内皮细胞[3H]-TdR掺入率较对照组和bFGF组明显升高,是对照组的11.12倍(P<0.05),bFGF组的4.05倍(P<0.05),表明串珠素和bFGF联合作用可以明显刺激内皮细胞增殖,这种联合作用较单独的串珠素或bFGF作用明显;PD98059组内皮细胞[3H]-TdR掺入率较缺氧+bF-GF组下降了93.40%(P<0.05),表明PD98059可以抑制串珠素与bFGF对内皮细胞的增殖作用。结论串珠素可以增强bFGF对心肌微血管内皮细胞的增殖作用,ERK介导了串珠素对bFGF的作用。
Objective To investigate the effects of pedecan on bFGF-induced proliferation of microvascular endothelial cells from the rat cardiac tissue. Methods Microvascular endothelial ceils(MEC) were isolated from the cardiac tissues of SD rats and cultured. The cultured cells were identified using immunocytochemistry assay with antibody for factor Ⅷ-related antigen and randomly distributed into experimental groups as follows: control group, hypoxic group, basic fibroblast growth factor(bFGF) group, hypoxia + bFGF group, perlecan antibody group and PD98059 group. The proliferation of MEC was detected using [^3H] -thymidine incorporation( [ ~ H] -TdR) assay. Results [^3H] -TdR of MEC in hypoxia + bFGF group was 11.12-fold and 4.05-fold respectively more than that of control group( P〈0.05) and bFGF group(P〈0.05). It meant that the interaction between perlecan and bFGF stimulated the MEC proliferation. [^3H]-TdR of MEC in PD98059 group reduced by 93.40% ( vs hypoxia + bFGF group, P〈0.05). It suggested that PD98059 inhibited the stimulatory effect of perlecan and bFGF on MEC proliferation. Conclusion Perlecan could promote the effect of bFGF on proliferation of microvascular endothelial cells from cardiac tissues. ERK mediated the effect of perlecan on bFGF.
