摘要
目的研究葛根素对胰岛素抵抗-高血压大鼠(IRH)的肿瘤坏死因子-α及肾素-血管紧张素的影响。方法用喂饲高糖高盐高脂的方法建立胰岛素抵抗-高血压(IRH)病理模型,随机分为模型卡托普利(Cap)组、葛根素注射液(Pue)高、中、低三个剂量组、模型阴性对照组,分别给药后,测定血浆胰岛素(Fins)、空腹血糖、肾素、血管紧张II(Ang-II)、肿瘤坏死因子(TNF-α)的含量及给药前后的血压变化。结果Cap和Pue80mg·kg-1及40mg·kg-1组均能明显降低IRH大鼠的肾素、Ang-II的含量及血压(P<0.05~0.01)。Pue20mg·kg-1组对上述各指标,则差异均未见显著性(P>0.05)。Cap和Pue80mg·kg-1组,能降低Fins及TNF-α含量(P<0.05~0.01),改善IRH大鼠的高胰岛素血症;提高低下的ISI,差异有高度显著性(P<0.01);而其余两个剂量组,则差异均未见显著性(P>0.05)。结论Pue可以有效增强胰岛素的敏感性,降低胰岛素抵抗-高血压大鼠的肾素—血管紧张素的含量,降低高血压。
Objective To study the effect of angiotension-Ⅱ in insulin resistant hypertensive (IRH) puerarin (Pue) on tumor necrosis iactoroand renninrats.Methods Feeding high fat-sugar-salt diets for 8 weeks to set up the model of IRH rats. And the rats were divided into 5 groups. The changes of blood pressure, contents of fasting blood glucose (FBG), fasting insulin (Fins), serum rennin, Ang-Ⅱ, TNF-αwere measured.Results Compared with the model control, groups of Captopril (Cap), Pue80mg.kg^-1 and 40mg.kg^-1 could significantly decrease the blood pressure, contents of rennin and Ang-Ⅱ (P〈0.05-0.01). Pue 20mg. kg^-1 hadn't significantly difference between the five groups. Cap and Pue 80 mg.kg^-1 could significantly decrease the Fins and TNF-α, improve the hyperinsulinmia, promote the insulin sensitivity index lowered (ISI) (P〈0.01) in IRH rats. And other two doses groups of Pue hadn't any difference in the three data (P〉0.05).Conclusion Pue can decrease blood pressure and enhance the insulin sensitivity, decrease the contents of rennin and angiotension Ⅱ in IRH rats.
