摘要
目的探讨骨髓间充质干细胞(mesenchymalstemcells,MSCs)对大鼠皮肤创面的促愈作用及机制,为临床皮肤损伤修复提供新的治疗方法。方法建立大鼠背部皮肤全层切割伤模型,将大鼠分为2组:空白对照组,MSCs处理组,分别于伤后3、6、9、12d采用大体观察,常规组织学检查观察2组大鼠背部皮肤伤口愈合状况及RT-PCR观察转化生长因子(TGF-β1)、表皮生长因子(EGF)mRNA在愈合中的变化特点。结果MSCs处理组大鼠皮肤愈合快,愈合指数高,创伤后第3天MSCs处理组大鼠皮肤TGF-β1上调明显,第6天EGF上调明显。结论MSCs能促进大鼠皮肤创面愈合,其机制可能与其引起各种生长因子的合成和分泌有关。
Objective To explore the effect of bone marrow mesenchymal stem cells (MSCs) on the repair of rat skin wounds, so as to provide a new method for clinical skin repair in the future. Methods A excision of 1 cm^2 full thickness wound were produced on the backs of 48 SD rats, and then the rats were randomly divided into saline control and MSCs treatment group. The progress of wound healing was observed on micro-and macro-levels at the 3, 6, 9, 12 d postinjury respectively. The expressions of transforming growth factor-β1 (TGF-β1) and epidermal growth factor (EGF) were detected by RT-PCR. Results MSCs treatment group showed faster and better wound healing. The MSCs treated group was also found to be regulated the expression of TGF-β1 and EGF in the wound tissues. Conclusion MSCs may improve the wound healing by promoting growth factor production and release in the skin wounds in rats, which may provide a foundation for the development of new treatment strategy for wound healing after skin injury.
