禽流感(H_9 亚型)HI稳定抗原的制备 被引量:2

摘要 禽流感(H9N2)灭活病毒液经浓缩纯化、离心及超声波处理,分组后加入不同物质如明胶、甘油、蔗糖作为稳定剂,制备HI诊断抗原。通过对不同温度条件(-20℃、4℃和室温)、不同存放时间诊断抗原HA效价的比较,筛选出了-20℃和室温保存6个月,4℃保存至少12个月抗原HA效价不发生改变的稳定剂———甘油。经实验室和田间试验,用该组抗原检测的禽流感H9亚型抗体与国家流感中心活病毒抗原检测结果基本一致。 Inactivited H9N2 AIV was treated by concentration, centrifugalization, and ultrasonic, then separately added with stabilizer: gelatin, glycerine and lactose. Then the antigen for HI was prepared which was deposited at -20℃, 4℃ and common temperature conditions. We detected HA titers in different temperature conditions and keeping times. The results showed that glycerine was the best stabilizer, and this group HI antigen can be kept for one year at least at 4℃, and 6 months at -20℃ and common temperature conditions with HA titer having no significant change. After the tests have been done in laboratory and fields, the detection results were conformed to the standard.
