摘要
目的:液相色谱-电喷雾离子阱串联质谱(LC~MS^n)联用法鉴别大鼠血浆中的阿托品及其主要代谢物。方法:取单剂量灌胃50mg·kg^(-1)阿托品的大鼠血样,甲醇沉淀蛋白,采用 LC-MS 及 LC-MS^n 等方法分析血样。和空白血样及阿托品相比较,根据血样中代谢物相对分子质量的变化(△M_r)及其多级质谱数据,鉴定并阐述其结构。结果:在服药后的大鼠血样中发现5种代谢物,分别为托品、N-去甲基托品、脱水阿托品、氧化阿托品以及托品酸.结论:该方法灵敏、快速、简便,适合于药物及其代谢物的快速鉴定。
Objective:To identify atropine and its metabolites in rat plasma by combining liquid chromatography and tandem mass spectrometry ( LC - MS^n). Methods: The plasma sampled from rats after single administration of 50 mg · kg^-1 atropine was added triple methanol to precipitate protein,then,it was analyzed by LC - MS^n. Identification and structural elucidation of the metabolites were performed by comparing their changes in relative molecular masses (ΔMt) ,retention - times and full scan MS^n spectra with those of the parent drug and blank plasma. Resuits :The results revealed that the parent drug and its five metabolites( tropine,N- demethyhropine, dehydrated atropine,hydroxyatropine and tropic acid) existed in rat plasma. Conclusion:This method is sensitive, rapid, simple, and it is suitable for the rapid identification of drug and its metabolites.
