α-硫辛酸对高糖诱导的系膜细胞核因子-κB活性的抑制作用 被引量:11

摘要 目的探讨α-硫辛酸对高糖诱导的大鼠系膜细胞增殖及核因子-κB(NF-κB)活性的影响。方法在体外条件下采用正常糖(NG)、高糖(HG)及HG+不同浓度α-硫辛酸处理大鼠系膜细胞;噻唑蓝法(MTT)测定系膜细胞增殖;采用凝胶电泳迁移率改变法检测NF-κB的活性。结果50~300μmol/L的α-硫辛酸可抑制系膜细胞增殖。HG刺激系膜细胞1h可引起NF-κB活性增强2.2倍(P〈0.01),高糖诱导的NF-κB的活化可被α-硫辛酸(200μmol/L)所抑制(0.6±0.3 vs 2.2±0.2,P〈0.01)。结论一定浓度的α-硫辛酸可抑制高糖诱导的系膜细胞增殖,并可降低NF-κB的DNA结合活性。 Objective To investigate the effects of α-lipoic acid on the proliferation and activation of NF-κB induced by high glucose (HG) in rat mesenteric cells (MCs). Methods The rat mesenteric cells were cultured in the medium with normal glucose (5.6mmol/L, NG), high glucose (25mmol/L, HG), HG+100 μmol/L α-lipoic acid, or HG+200μmol/L α-lipoic acid and HG+PDTC (a NF-κB inhibitor). Activation of nuclear factor-κB (NF-κB) of rat mesenteric cells were measured by electrophoretic mobility shift assay (EMSA). The cell proliferation was assessed by MTT. Results α-lipoic acid (50~300μmol/L) can inhibit the proliferation of MCs. The NF-κB binding activity was 2.2-fold higher in MCs exposed to HG compared to NG (P〈0.01). When the MCs were cultured in the presence of α-lipoic acid (200μmol/L) for 1h, there was a highly significant reduction in NF-κB binding activity (0.6±0.3 vs 2.2±0.2, P〈0.01). Conclusion It is suggested that α-lipoic acid may inhibit high glucose-induced NF-κB activation in mesenteric cells. These findings may provide an experimental evidence for further evaluating the possible protective effect of α-lipoic acid against diabetic nephropathy.
