嗅球切除对成年大鼠侧脑室外侧壁神经生发活动的影响 被引量:1

THE EFFECTS OF OLFACTORY BULB REMOVAL ON NEUROGENESIS IN THE SUBVENTRICULAR ZONE IN ADULT RAT BRAIN

摘要 目的研究成年大鼠嗅球切除后侧脑室外侧壁(SVZ)的形态学变化,探讨嗅球对成年大鼠SVZ神经生发活动的影响。方法建立成年SD雄性大鼠右侧嗅球切除模型,并分别存活2、4、8、12周;利用Nissl染色、多唾液酸神经细胞黏附分子(PSA-NCAM)、GFAP免疫组织化学染色的方法,分别观察成年SD大鼠嗅球切除后存活不同时间两侧SVZ的组织结构和免疫组织化学特征;计数模型动物脑两侧SVZ细胞总数及PSA-NCAM、GFAP免疫阳性细胞数,并进行统计学分析。结果嗅球切除4周后,嗅球切除侧SVZ的细胞总数增加,PSA-NCAM免疫阳性细胞数增加,但GFAP免疫阳性细胞数没有明显变化;SVZ的细胞总数及PSA-NCAM免疫阳性细胞数的增加有从SVZ嘴侧向尾侧蔓延的趋势。结论嗅球切除后在SVZ仍有新生神经元不断产生,说明SVZ的神经生发活动可能并不依赖于嗅球的存在。 Objective To investigate the morphological change of olfactory bulb(OB) removal in the subventricular zone (SVZ) and explore the effect of OB on the neurogenesis in the SVZ in adult mammals, and to provide more data to know better about the mechanism of neurogenesis in that area. Methods 1. An animal model of OB removal was created and the animals survived 2,4,8,12 weeks respectively. 2. The Nissl staining and immunohistocbemistry for glial fibrillary acidic protein (GFAP), polysialylated form of the neural cell adhesion molecule (PSA-NCAM) were used in the present study and observed under lightmicroscope. 3. Total cell number and the cell number of PSA-NCAM and GFAP-positive in two side of SVZ in the animal models were counted and analyzed. Results 1. The total cell number and the PSA-NCAM-positive cell number in the operation side of the SVZ in the rats of OB removal increased than that of the controlateral one, and the GFAP-positive cell number did not change. 2. Increase of the cell number extended gradually from the restral SVZ toward the caudal. Conclusion Young neurons are still produced in the SVZ after the OB removal, suggesting that OB is not essential for neurogenesis in the SVZ.
