HIF-1α的表达与脑膜瘤细胞增殖的关系

摘要 目的:探讨缺氧诱导因子-1α的表达与脑膜瘤细胞增殖的关系。方法:采用免疫组织化学方法检测48例脑膜瘤和10例正常脑膜组织中HIF-1α的表达,并通过检测增殖细胞核抗原(PCNA)的表达来反映肿瘤细胞的增殖活性,对所得资料进行统计学分析。结果:48例脑膜瘤组织中,HIF-1α阳性表达率为79%,其中弱阳性(+),阳性(++),强阳性(+++)表达率分别为33%,31%,15%;PCNA-LI为1-5,平均为1.8±0.7;正常脑膜中无HIF-1α、PCNA表达,PCNA-LI为1,明显低于肿瘤组织,差异有显著性(P〈0.01)。HIF-1α阴性表达者,PCNA为1.2±0.6;呈弱(+)、中(++)及强阳性(+++)表达者,PCNA分别为1.9±0.7、2.7±1.1及3.6±1.3,HIF-1α阳性表达程度与PCNA-LI呈正相关(ra=0.726,P〈0.01)。结论:HIF-1α的表达与脑膜瘤细胞增殖密切相关,进一步研究HIF-1α在脑膜瘤中的表达及调控血管生成细胞增殖的机制,可能为脑膜瘤的治疗开辟新方法。 Objective: To study the relationship of HIF-1α expression and proliferative activity in meningioma. Methods: Forty-eight meningioma tissues and ten human normal meningeal tissues maters expressions were detected for HIF-1α expression by SABC immunohistochemical method and the PCNA by SP immunohistochemieal method were used to label proliferative activity. The PCNA-labeling index(PCNA-LI) of tumors were calculated. Results: The positive expression rate of HIF-1α was 79% in 48 tumor tissues, among which the weak, moderate and strong positive expression rate were 33%, 31% and 15% respectively; The mean (x^-± s) of PCNA-LI was 1.8 ± 0. 7 ( ranging from 1 to 5 ) ; otherwise the expressions of HIF-1α and PCNA in normal meningeal tissues were negative, the PCNA-LI was 1. The differences of HIF-1α and PCNA-LI between meningioma tissues and human normal meninges were significant(P 〈0. 01 ). The PCNA-LI in the negative, weak, moderate and strong HIF-1α expression samples were 1.2 ±0. 6, 1.9 ± 0. 7, 2.7 ±1.1 and 3.6 ±1.3 respectively, PCNA-LI was positively correlated with HIF-1α expression(ra = 0. 726, P 〈 0. 01 ). Conclusion: The expression of HIF-1α in meningioma is significantly correlated to tumors' proliferative activity; further studying the mechanism of the HIF-1α expression and proliferative activity would lead to new ways to treat meningiomas.
