丹芍化纤胶囊对大鼠肝纤维化的预防作用及对HSCs增殖和活化的影响

The Preventive Actions of Traditional Chinese Medicine DanShaoHuaXian Capsules on Rat Liver Fibrosis Induced by Pig Serum and the Effects of DanShaoHuaXian Capsules on Proliferation and Activation of Hepatic Stellate Cells

目的:探讨中药复方制剂丹芍化纤胶囊对大鼠免疫性肝纤维化的预防作用及其对肝星状细胞增殖、活化的影响。方法:W istar雄性大鼠分为正常组、病理模型组、丹芍化纤胶囊低剂量组(0.3g/kg)、中剂量组(0.6g/kg)、高剂量组(0.9g/kg)及秋水仙碱组(0.15mg/kg)。除正常组外,各组均腹腔内注射猪血清(0.5mL只/,2次/周,持续12周)诱导大鼠免疫性肝纤维化模型,并同时灌胃给予相应药物,持续12周。实验第8、10周检测尿羟脯氨酸排出情况。大鼠肝脏常规病理切片行M asson三色法观察纤维化程度。肝脏免疫组织化学SABC法染色结蛋白(desm in)及α-平滑肌肌动蛋白(α-SMA)观察肝星状细胞(HSCs)的增殖、活化。结果:丹芍化纤胶囊中、高剂量给药能增加大鼠尿羟脯氨酸的排出量(P<0.01),抑制HSCs的增殖(P<0.01),减少活化HSCs的数量(P<0.01),减轻肝纤维化程度。结论:丹芍化纤胶囊能减轻肝脏损伤,增加胶原降解,抑制HSCs的增殖和活化,具有预防大鼠肝纤维化的作用。

Objective： To investigate the preventive actions of DanShaoHuaXian capsules,a traditional Chinese medicine, on rat liver fibrosis and the effects of DanShaoHuaXian capsules on proliferation and activation of hepatic stellate cells in fibrotic rat livers induced by pig serum. Methods： Male Wistar rats were derided into 6 groups. Except for the normal group, the other five groups were given an intraperitoneal injection of pig serum （0.5mL, twice a week for 12 weeks） to make the rat liver fibrosis. DanShaoHuaXian capsules prevention groups were given DanShaoHuaXian capsules （0.3.0.6.0.9g/kg, P. O, daily for 12 weeks） simultaneously to examine the protective actions. Colehicine group were administrated Colchicine （0. 15mg/kg, P. O, daily for 12 weeks）. The urinary excretion of hydroxyproline was detected at 8^th and 10^th week. The experimental rats were killed at the end of the 12^th week. Liver samples were histopathologically analyzed by Masson stain. Proliferation and activation of hepatic stellate cells （HSCs） were stained by inmunohistochemistry to detect the desmin and α - smooth muscle actin（ α - SMA） postive cells. Results： DanShaoHuaXian capsules improved collagen degradation （ P 〈 0.01 ）, inhibited proliferation of HSCs （ P 〈 0.01 ） , decreased the number of activated HSCs（ P 〈 0.01 ）, alleviated liver fibrosis. Conclusion ：DanShaoHuaXian capsules shows good effects on the prevention of rat live fibrosis induced by pig serum. The effects may be related to improving collagen degradation ,inhibiting proliferation and activation of HSCs.
