摘要
目的:建立HPLC-ELSD法测定川麦冬须根大孔树脂富集物中麦冬皂苷D、D′的含量。方法:以麦冬皂苷D、D′为对照品,应用HPLC-ELSD法,色谱柱:APOLLO C18柱(100A,5μ,4.6mm×250mm),流动相:乙腈-水(50:50);流速:1.0ml/mi。;柱温:25℃;检测器:蒸发光散射检测器(ELSD),雾化温度:70℃,蒸发温度:90℃,气体流速:1.5SLM。结果:麦冬皂苷D在4.424。22.12μg范围内线性良好,相关系数r=0.9997,加样回收率为:97.6%,RSD=1.51%;麦冬皂苷D′在3.848。19.24μg范围内线性良好,相关系数r=0.9998,加样回收率:98.4%,RSD=4.80%。结论:本方法线性关系良好,重现性佳。
AIM: To establish a HPLC-ELSD method for determining ophiopogonin D and ophiopogonin D′ in Fibre Ophiopogon enriched extract by macroporous resin. METHODS: The determination was performed by HPLC-ELSD on APOLLO C18 column (100 A,5μ,4.6 mm×250 mm) ,using acetonitrile-H2O(50: 50) as a mobile phase, flow rate was at 1.0 mL/min, column temperature was at 25℃, detector: ELSD ( Neu : 70℃, Eva : 90℃, gas flow : 1.5 SLM). RESULTS : The linear range of ophiopogonin D was 4.424-22.12μg, r = 0. 999 7, The average recovery was 97.6% ,RSD = 1.51%. The linear range of ophiopogonin D′ was 3. 848-19.24μg,r= 0. 999 8. The average recovery was 98.4% ,RSD = 4.8%. CONCLUSION: The method is convenient, sensitive and accurate.
