二烯丙三硫对二甲基肼引起小鼠致突作用影响

摘要 为探讨大肠癌的发生机理和防治机制,选用二甲基肼作为昆明种小鼠大肠致突(癌)物,将二烯丙三硫作为阻断剂,应用大肠隐窝上皮细胞核异常检测法和嗜银核仁形成区检测法技术,观察二烯丙三硫对二甲基肼引起大肠癌早期突变作用的影响。实验证明:用二烯丙三流处理组小鼠与单纯用二甲基肼处理组小鼠的隐窝上皮细胞核异常和核仁形成区变化相比较均有显著性差异(P<0.01),其核仁形成区的形态上也有显著不同。表明二烯丙三硫在二甲基肼引起小鼠大肠癌变的早期致突作用时起到抑制作用,同时提示大肠隐窝上皮细胞核异常的检测可用于检测致突(癌)物和抑制物,而核仁形成区的观察亦可反映化学物质对染色体的损伤或修复所起的作用。 We selected DMH as mutagen to Kunming mice and DATS as obstructive agent, studied the genetic and proventive mechanism of colonic carcinoma. Colonic epithelial nuclear aberration bioassay and nucleolar organizer region (NOR) method were applied in our experiment for observing the response of DATS in early mutation of colonic carcinoma induced by DMH.The result showed it was significant that the DATS-group was far lower than the DMHgroup in colonic epithelial nulear aberrations and NORs (P<0. 01 ). The result suggested that DATS might obstruct the early mutation of epithelial cell induced by DMH in detecting mutagen and obstructive agent. The NOR method might be used in reflection on the injured and repairable of chromosome by chamical material.
