摘要
目的:探讨尿多酸肽对外周血辅助性T1淋巴细胞(Th1)分泌白细胞介素-2(IL-2)和干扰素-γ(IFN-γ)的影响。方法:选择肿瘤患者和健康查体者各30例,抽取静脉血,分离外周血单个核细胞(PBMCs),用10%胎牛血清1640培养液将其浓度调成1×106个·mL-1,分別加入不同浓度的尿多酸肽注射液,培养72 h后,收集细胞悬液-70℃冻存,经反复冻融、离心收集上清液,用酶联免疫吸附法(ELISA)测定上清液中IL-2和IFN-γ的含量。结果:在加入不同浓度(0,0.1,0.2.0.4 mg·mL-1)的屎多酸肽培养后,肿瘤患者PBMCs上清液中IL-2含量分别为(91.10±35.80),(40.25±14.38),(33.40±13.42)和(21.35±9.58)pg·mL-1,健康人分别为(127.76±47.91), (73.43±29.77).(51.62±19.85)和(34.78±14.95)pg·mL-1;肿瘤患者PBMCs上清液中IFN-含量分别为(183.38±66.75),(107.48±40.33),(79.86±31.03)和(52.02±22.67)pg·mL-1,健康人分别为(249.45±89.69),(160.12±62.08),(105.04±39.47)和(69.58±26.25)pg·mL-1。在相同尿多酸肽浓度下,患者的IL-2和IFN-γ含量低于健康人,且有统计学差异(P<0.05);随培养液中尿多酸馱浓度的增加,肿瘤患者和健康人的IL-2和IFN-γ的含量逐渐降低,且有显著差异(P<0.05)。结论:肿瘤患者的Th1细胞的免疫功能低于健康人;
Objective: To analyze possible roles of variable concentrations of uroacitides for injection on the cytokines levels mediated by the helper T1 lymphocytes (Th1). Methods : The peripheral blood mononuelear cells (PBMCs) were extracted from the blood samples of tumor patients and healthy volunteers (30 subjects each). The PBMCs were plated at a density of 106 cells·mL^-1 in RPMI 1640 and then mixed with uroacitides for injection of different concentrations. After incubated for 72 hours, the PBMCs were harvested and stored at -70 ℃. After repeatedly frozen and thawed, the incubated PBMCs were centrifuged and the supernatant was gathered. The concentrations of interleukin-2 (IL-2) and interferon-γ(IFN-γ) in the supernatant were assayed by ELISA. Results: After incubation with different concentration of uroactides for injection (0,0. 1,0.2,0.4 mg·mL^-1 ), the IL-2 concentrations in the supernants of the patients' and healthy volunteers' PBMCs were (91.10 ± 35.80) vs. ( 127.76 ± 47.91), (40.25 ±14.38)vs.(73.43±29.77), (33.40±13.42)vs.(51.62±19.85) and (21.35 ± 9.58)vs. (34.78 ± 14.95) pg·mL^-1, respectively. The concentrations of IFN-γ were ( 183.38 ± 66.75 ) vs. (249.45 ± 89.69 ), ( 107.48 ± 40. 33 ) vs. ( 160. 12 ± 62. 08 ), (79.86 ± 31.03 ) vs. ( 105.04 ± 39, 47) and (52.02 ± 22.67 ) vs. (69.58 ± 26.25 ) pg·mL^-1, respectively ; the P values showed the statistical difference (P 〈 0.05 ). The IL-2 and IFN-γ levels were inversely proportioned to the increased concentrations of uroacitides for injection (P 〈 0.05 ). Conclusion: The tumor patients deteriorate the cell-mediated immunity function of Thl cells more than the healthy volunteers. Uroacitides inhibits the cytokine secretion mediated by Thl cells in vitro.
