摘要 目的研究地塞米松和辛伐他汀对成骨细胞增殖分化的影响。方法从新生SD大鼠颅盖骨分离培养成骨细胞,随机分为3组培养,A组加入1×10-7mol/L的地塞米松、1×10-7mol/L的辛伐他汀和二甲基亚砜(DM-SO,终浓度为1g/L);B组加入1×10-7mol/L的地塞米松和与A组等量的DMSO;C组只加入与A组等量的DMSO作为对照。96h后,以RT-PCR一步法分析成骨细胞骨钙素mRNA的表达;采用MTT法测定成骨细胞增殖率。结果A、C组成骨细胞骨钙素mRNA表达量及细胞增殖率均高于B组(P<0.01),A与C组比较差异无统计学意义(P>0.05)。结论辛伐他汀可对抗地塞米松对成骨细胞增殖和分化的抑制作用。 Aim : To study the effect of dexamethasone and simvastatin on the osteoblast proliferation and differentiation. Methods: Osteoblasts were separated from skull of newborn SD rats, and randomly divided into 3 groups (8 samples for each group). Group A was fed with culture medium RPMI 1640,and treated with simvastatin of 10^-7 mol/L, dexamethasone of 10^-7 mol/L and DMSO of 0. 1% ;Group B was maintaioed in culture medium RPMI 1640 and treated with dexamethasone of 10^ -7 mol/L and DMSO of 0. 1% ;Group C was only cuhured with DMSO of 0. 1%. After 96 hours, the expression of osteocalcin mRNA was observed by RT-PCR and the proliferation of osteoblast was measured using MTT reduction assay. Results: The expression of osteocalcin mRNA and the rate of proliferation of osteoblast of Group A and C were significantly higher than those of group B (P 〈 0.01 ), and there was no significant difference between group A and group C ( P 〉 0. 05 ). Conclusion: Simvastatin is resistant to the restraint of the osteoblast proliferation and differentiation by dexamethasone.
