摘要
目的观察乙醇对人骨髓间充质干细胞(hBMSCs)内成脂转录因子(PPAR)γ和骨钙素(Osteocalcin)mR-NA表达的影响。方法取健康自愿者骨髓,通过梯度离心、贴壁分离培养,获得hBMSCs,传至第2代hBMSCs8d,随机分为2组,实验组以0.09mol/L乙醇作为诱导剂每d处理细胞,对照组不给予乙醇,继续培养细胞6d。应用RT-PCR技术检测2组细胞中PPARγmRNA和OsteocalcinmRNA的表达。结果与对照组比较,实验组hBMSCs内PPARγmRNA表达增高(1.38±0.27vs1.73±0.34,P<0.05)。与对照组比较,实验组hBMSCs内OsteocalcinmR-NA表达降低(1.08±0.22vs0.48±0.11,P<0.05)。结论乙醇能够诱导hBMSCsPPARγ表达增强,Osteocalcin表达减少,这可能与酒精性骨坏死的发生机制有关。
Aim : To observe the effect of alcohol on the expression of adipogenic transcription factor (PPAR)γ mRNA and osteocalcin mRNA in human bone marrow mesenchymal stem cells (hBMSCs). Methods:Bone marrow was aspirated from the healthy adult human, and then the cells were cultured in vitro. The hBMSCs were isolated and obtained after the first passage, and then were divided into two groups. The cells were treated with or without 0.09 mol/L alcohol. The ex- pression of PPAR3, mRNA and osteocalcin mRNA in the hBMSCs of two groups was detected by RT-PCR technique. Results: The expression of PPAR3, mRNA in the cells treated with alcohol (1.73± 0.34) was significantlly higher than that in the cells treated without alcohol (1.38± 0.27 ) (P 〈 0.05 ). The expression of osteocalcin mRNA in the cells treated with alcohol (0.48 ±0. 11 ) was significantly lower than that in the cells treated without alcohol ( 1.08 ±0.22) ( P 〈 0.05). Conclusion: Alcohol could induce higher PPAR'v expression and lower osteocalcin expression of hBMSCs, which may inw)lve in the alcohol-induced osteonecrosis.
