摘要
目的探讨银杏叶提取物制剂EGb 761对大鼠视神经损伤后视网膜神经节细胞(RGC)存活的影响。方法大鼠48只经荧光金逆行标记后制备视神经损伤的动物模型,随机分为治疗组和对照组,两组分别给予EGb 761 150 mg/kg.d和生理盐水灌胃。在视神经损伤后4d、7d及14d观察视网膜铺片,进行RGC计数。结果大鼠视神经损伤后4d、7d及14d,RGC数量持续减少,但治疗组均高于对照组,各时间点的差异均有统计学意义(依次为P<0.05,P<0.01,P<0.01)。结论EGb 761可促进视神经损伤后RGC的存活,对RGC有保护作用。
Objective To investigate the effect of EGb 761 on the survival of injured retinal ganglion cells (RGC) in an optic nerve crush model in adult rats. Methods Healthy adult Sprague Dawley rats were divided randomly into control group and treating group. Seven days after the injection of Flurogold into Superior Colliculous, unilateral crush of the optic nerve was performed. Operated animals received gayage of EGb761 (150mg/kg·d) or vehicle immediately after crush. All animals were sacrificed 4,7 and 14 days after optic nerve crush respectively. Flurogold-labled retinal wholemounts were studied and the number of RGC was calculated. Results Treatment with EGb761 significantly augmented RGC survival after optic nerve crush, with more RGC observable in treating group than in control group (P 〈 0. 05, P 〈 0.01 London, P 〈 0.01 ) Conclusions This study suggests that EGb761 has a definite protective effect on RGC after optic nerve crush, which make it to be a potential neuroprotective agent for injured RGC.
