摘要
目的探讨核因子κB(NF-κB,p65)在胰腺癌组织中的表达及其与VEGF、p53蛋白表达的相互关系。方法电泳迁移率变动分析(EMSA)测定45例胰腺癌及癌旁组织、8例慢性胰腺炎和9例正常胰腺组织中NF-κB的活性,Western印迹法检测NF-κB、IκB-a蛋白表达;免疫组织化学法检测胰腺癌中VEGF、p53、p65蛋白表达。结果NF-κB在肿瘤组织中被激活;NF-κB在胰腺癌、旁组织、慢性胰腺炎、正常胰腺组织中阳性表达率分别为71.0%、20.0%、12.5%、0(P<0.05);NF-κB基因表达与胰腺癌的分化程度、临床分期、肿瘤大小无关(P>0.05),与淋巴结转移和周围浸润有相关性(P<0.05);转移癌中NF-κB蛋白表达与VEGF、p53蛋白表达显著相关。结论核因子κB基因在胰腺癌中过度表达并与VEGF、p53一起参与胰腺癌的浸润、转移过程。
Objective To study the expression of nuclear factor kappab gene(NF-κB) and its relationship with expression of VEGF,p53 proteins in pancreatic cancer(PC). Methods The expression of NF-κB, IκB-a was investigated by Western blot in PC samples, parocarcinoma tissues, chronic pancreatitis (CP) samples and normal pancreatic(NP) samples. The activity of NF-κB was detected through eletrophoretic mobility shift assay(EMSA) in PC, CP and NP samples. Immunohistochemical analysis for VEGF, p65, p53. Results The positive expression rate of NF-κB protein determined by Western-Blotting analysis is 71% in PC tissues,20% in parocarcinoma tissues,and 12. 5% in CP,0/9 in NP, there was significant difference between the 4 groups(P〈0. 05). There was no relationship between NF-κB gene expression and histological diffentiation, clinical stages(P〉0. 05 ), but the lymph node metastasis was ( P〈0. 05 ), the expression of NF-κB gene correlated with VEGF, p53 expression. Conclusion NF-κB may play an important role in pathgenesis of PC. Upregulated NF-κB and disfunction of VEGF,p53 may play a synergetic role in the process of carcinogenesis of PC.
