摘要
目的观察灯盏花素对大鼠全脑缺血再灌注后海马神经元L型钙通道平均开放概率和开放时间的影响。方法制作全脑缺血大鼠模型,再灌注1.5 h、3.0 h、4.5 h和6.0 h,急性分离海马CA1区神经元,采用膜片钳技术细胞贴附式方法记录神经细胞膜上单通道Ca^(2+)电流,分析L型钙通道平均开放概率和开放时间,在此基础上观察灯盏花素对其影响。结果不同浓度的灯盏花素能降低海马神经元L型钙通道的平均开放概率和开放时间,且在缺血再灌注3.0 h内作用明显。结论灯盏花素对脑缺血再灌注脑损伤有一定的保护作用。
Objective To observe the effect of breviscapine on the open probability (Po) and the average open time (To) of L -type calcium channel in the hippocampal neurons of the whole brain ischemia -reperfusion rat model. Methods The whole brain ischemia rat model was established. At the reperfusion periods of 1.5 h, 3.0 h, 4. 5 h and 6.0 h, the hippocampal CA1 neurons were dissociated abruptly and the Ca2~ electricity of single channel on the neuron membrane were recorded with cell - attach model of patch clamp techniques. The Po and To of the L - type calcium channel were analyzed and the effect of the breviscapine was observed. Results Different concentrations of breviscapine can decrease the Po and To of the L - type calcium channel in hippocampal neurons, particularly within 3 hours after ischemia - reperfusion. Conclusions Breviscapine has protective effect on the cerebral ischemia - reperfusion injury.
