白介素-8基因与早期糖尿病大血管并发症的关系 被引量:4

Relationship between IL-8 gene expression and vascular complications in early diabetes mellitus

摘要 目的研究早期2型糖尿病(T2DM)患者白介素-8的变化。方法随机选择无并发症新诊断的T2DM患者,应用酶联免疫吸附法(ELISA)和逆转录-聚合酶链反应(RT-PCR)技术检测白介素-8(IL-8)mRNA转录半定量水平和蛋白表达;并观察T2DM组患者口服75g葡萄糖2小时后IL-8的变化;分析血糖以及血浆真胰岛素水平对IL-8的影响。结果IL-8mRNA半定量均明显高于健康对照组,而两组血浆中IL-8浓度差异无统计学意义;服糖后血糖与IL-8mRNA呈相关性。T2DM患者IL-8与血浆真胰岛素水平无相关性。结论单纯血糖升高可以引起IL-8介导单核细胞的粘附及通过内皮细胞这一过程,血糖可能为独立的动脉粥样硬化相关因子。 Objective To investigate the changes of interleukin-8(IL-8) in the patients with car ly type 2 diabetes mellitus(T2DM). Methods To study the first-diagnosed T2DM patients withoul complications and normal group; mRNA and protein expression of interleukin-8 were measured by ELISA and RT-PCR. Subsequently, the relationship between plasma glucose or real-active insulin and interleukin 8 was analysed. Results Semi-quanthatlve mRNA of IL-8 DM group was significantly higher than that of the mormal group. There was no significant difference in IL-8 concentration between the two groups. The fasting glycaemia associated with IL-8 level, but there was no correlation between the concentration of insulin and IL-8. Conclusion Hyperglycemia may induce IL-8 mediated monocyte adhesion and process of filtering into vessel endothelium; glyeaemia maybe an independent risk factor to atheroselerosis.
