摘要
目的探讨血清饥饿条件下成骨细胞的增殖及胰岛素样生长因子-Ⅰ(IGF-Ⅰ)的作用。方法采用组织块培养法体外培养成骨细胞,分正常血清浓度组,血清饥饿组,IGF-Ⅰ组进行干预,观察细胞生长状态,MTT法检测细胞增殖。结果血清饥饿组成骨细胞贴壁减少,融合缓慢,增殖低于正常血清浓度组(P<0.01),IGF-Ⅰ组细胞增殖较血清饥饿组增加(P<0.01),低于正常血清浓度组(P<0.01)。结论血清饥饿抑制成骨细胞增殖,可能是缺血性骨病的发病机制之一。IGF-Ⅰ能部分改善由于血清饥饿引起的成骨细胞抑制。
[Obieetive] To study the proliferation of osteoblasts under serum starvation and the effect of insulin-like growth factor-I (IGF-I) on it. [Methods]Osteoblasts were cultured in vitro with different inter ventions. The growth of osteoblasts was observed under microscppy and detected with MTT assay. [Results] The proliferation of osteoblasts cultured under serum starvation was inhibited ( P 〈0, 01 ), and improved by IGF-I ( P 〈0.01). [Conclusion] Osteoblastic proliferation is inhibited by serum starvation, which may be one of mechanisms of avascular osteonecrosis. IGF- I can partially prevent osteoblasts inhibition induced by serum starvation.
