摘要
目的观察关节炎疼痛对大鼠脊髓促炎性细胞因子IL-1β和TNF-α水平的影响。方法Wistar大鼠20只,150~180g,雌雄各半,随机分为对照组和实验组。对照组右后踝关节腔注射生理盐水,实验组左后踝关节腔注射完全弗氏佐剂,制备单发佐剂性关节炎动物模型。4周后用屈关节疼痛试验评分法测定关节炎疼痛行为学变化,取腰段脊髓,ELISA法测定脊髓中IL-1β和TNF-α含量。结果实验组屈关节疼痛试验评分较对照组明显降低(P<0.01),IL-1β和TNF-α水平较对照组明显升高(P<0.01)。结论关节炎疼痛时脊髓IL-1β和TNF-α水平升高,提示脊髓促炎性细胞因子与关节炎疼痛的病理生理机制有关。
[Objective] To investigate effect of arthritic pain on the levels of proinflammatory cytokine IL-1β and TNF-α in the lumbar spinal cord of arthritis rats. [Methods] 20 Wistar rats were randomly divided into control group (n =10) and experimental group (n =10). The arthritis model was done by administered CFA into joint cavity of the rats in experimental group. Saline was administered into joint cavity of rats in the control group. Pain behaviors, i.e. the leg-withdrawal response scores, were measured with arthritic flexion pain test four weeks after CFA. The levels of IL-1β and TNF-α in lumbar spinal cord the fourth week after adjuvant-induced arthritis. [Result] The legwithdrawal scores in inflammed leg were significantly lower in experiment group than that in control group (P 〈0.01). Both IL-1β and TNF-α in the lumbar spinal cord were higher signifieandy in experiment group than those in control group (P 〈0.01). [Conclusion] IL-1β and TNF-α in spinal cord of the rats in adjuvant-induced arthritis were markedly increased. It is suggested that there is a correlation between IL-1β and TNF-α in spinal cord and chronic arthritic pain.
