摘要
对螺旋藻藻蓝蛋白的提取和纯化方法进行了研究和优化,发现用pH=7.0、0.067mol/L的磷酸盐缓冲液粗提取处理,再用50%饱和度硫酸铵盐析纯化所得到的藻蓝蛋白的纯度和提取量都比较理想。在实验条件下纯度和提取率呈负相关,其中盐析后藻蓝蛋白的纯度达A620/A280=0.508,提取率W为1.12%。将样品进行了红外光谱分析的定性确认,其荧光光谱的室温荧光发射峰、紫外吸收特征峰与文献值有较好的吻合。
Take off to Study and optimize On the method of the Extraction and Putifiecation of Phycocyanobily faau Spirulina Platensis, discover to use the buffer solution of phosphoric acid salt handling, ( pH = 7.0, 0. 067mol/L), the pure degree and pick up quantity of Phycocyanobily both satisfied outcome.No matter what roughing or salt out for Purifiecation with (NH4)2SO4 solution of 50% saturation degree. Each other of pure degree and pick up quantity presented negative interrelation in the experiment term. The pure degree of Phycocyanobily in salt out reaches the A620/A280 = 0. 508, withdraw rate W as 1.12 %. The sample obtained qualitatively confirmation by FT- IR spectrun . Its Fluorescence and UV spectrum absorption characteristic date were compatiblc with those of the literature.
